Selective Enrichment Broth That Promotes The Efficient Repair And Growth Of Heat –And Freeze- Injured Escherichia coli O157:H7 Comparable To A Non-Selective Broth

Broth Function

The purpose of an enrichment broth is to amplify very low levels of a specific bacterium within 24 hours so it can be more readily identified and isolated by a subsequent methodology. In situations where cells of the targeted bacterium are very low and injury is high, such cells must undergo repair without deleterious effects. The broth must also contain selective agents to suppress background growth which can interfere with the isolation of the target bacterium. Our selective E. coli O157:H7 enrichment broth permits the proliferation of both heat- and freeze- injured and non-injured E. coli O157:H7 strains while inhibiting the growth of Gram (+) and selected Gram (-) bacteria.

Advantages/Benefits

1. Promotes heat– and freeze- injury repair in E. coli O157:H7 at both 37 and 42°C comparable to the non-selective Tryptic soy broth (TSB).

2. Heat– and freeze- injury repair is superior to the buffered peptone water supplement with vancomycin, cefsulodin and cefixime broth (BPW-VCC).

3. Supports growth rates and cell densities of unstressed E. coli O157:H7 cells comparable to growth in TSB and superior to BPW-VCC.

4. Can be used as an enrichment broth for E. coli O157:H7 in conjunction with subsequent culture methods or other techniques such as PCR.

5. Allows shorter enrichment times for both heat– and freeze- injured cells at very low levels (1-2 cells/25 g sample) of E. coli O157:H7 than the BPW-VCC and the modified EC plus novobiocin before identifying presumptive positives by PCR.

6. Ease of preparation with forty-five day stability of broth stored at 2-8°C in the dark.

7. One bottle of broth powder and one box of supplements will make approximately 12.5 liters of enrichment broth.
