1. Our broth supported the growth of 28 non-O157 STEC strains (which included the 6 major serogroups) with a similar final cells density as cells grown in Tryptic Soy Broth (TSB), whereas the USDA listed modified Tryptone Soya Broth (m-TSB) inhibited the growth of more than 20% of these strains incubated at 41-42°C for 20-22 hours.
2. In our broth only a portion of the freeze-injured non-O157 STEC cells are killed without affecting the survival of the non-injured cell population, whereas, m-TSB killed the freeze-injured non-O157 STEC cells and a substantial portion of the non-injured cells.
3. Our broth, therefore, is more effective than m-TSB in supporting the growth and survival of freeze-injured non-O157 STEC cells incubated at 41-42°C.
4. The selectivity of our broth and m-TSB is similar; however, our broth inhibits the growth of pseudomonad type bacteria which can be the dominate organism in fresh meats and m-TSB does not.
5. In our broth, as in the nonselective TSB broth, the growth of freeze-injured non-O157 STEC cells is initiated after 2 hours of incubation at 41-42°C.
6. Ease of preparation with forty-five day stability of broth stored at 2-8°C in the dark.
7. One bottle of broth powder and one box of supplements will make approximately 12.1 liters of enrichment broth.
