Colony Appearance

Presumptive positive colonies of Yersinia pestis appear as blue-black raised colonies 0.5-2.0 mm in diameter with or without colorless halos after 48-72 h at 26-30°C. Y. enterocolitica, Y. pseudotuberculosis, Y. kristensenii, Y. intermedia and other Yersinia species will grow as green to yellow colonies 1.0-2.0 mm in diameter. Other Gram (-)s either are inhibited or grow as green, blue-green, yellow or white colonies.
Gram (+)s are inhibited.

Mechanism

The Yersinia pestis medium uses the chromogenic substrate X-β-D-glucopyranoside to detect the enzyme β-glucosidase which is found in all Y. pestis strains. In addition, this medium contains sucrose, L-rhamnose, cellobiose, and lactose which Y. pestis cannot ferment. Therefore, the Y. pestis produces a blue-black colony in the presence of the pH indicator brom cresol purple, whereas other Yersina species which ferment one of the sugars produce green to yellow colored colonies.
